結果和討論

溶(rong)(rong)解度是一個常(chang)見(jian)且經(jing)常(chang)被忽略的(de)參數,難以測(ce)量,甚至更難理解。由于(yu)強大(da)的(de)晶體(ti)(ti)力(li)(li),固體(ti)(ti)可(ke)能(neng)(neng)不溶(rong)(rong)于(yu)水,即使有X射(she)線數據,晶體(ti)(ti)力(li)(li)的(de)性(xing)質也常(chang)常(chang)模糊不清。在(zai)我(wo)們的(de)例子(zi)中,含有酰胺的(de)分(fen)子(zi)內(nei)和分(fen)子(zi)間(jian)氫鍵可(ke)能(neng)(neng)有助(zhu)于(yu)固態的(de)熱力(li)(li)學穩(wen)定性(xing)。不溶(rong)(rong)性(xing)也可(ke)能(neng)(neng)源于(yu)溶(rong)(rong)液中不利的(de)溶(rong)(rong)劑化和聚集(ji)效應。A系列海藻糖雙(shuang)子(zi)可(ke)能(neng)(neng)會抵抗溶(rong)(rong)解,因(yin)為(wei)它們無法以與相應單(dan)鏈(lian)段可(ke)能(neng)(neng)的(de)方式形成親(qin)水膠(jiao)束聚集(ji)體(ti)(ti)。當考慮到(dao)雙(shuang)子(zi)座的(de)兩條(tiao)鏈(lian)位于(yu)雙(shuang)糖的(de)遠端,分(fen)子(zi)內(nei)鏈(lian)/鏈(lian)相互作用部分(fen)受損時(shi),這種推(tui)測(ce)就(jiu)變得可(ke)信了(le)。鏈(lian)關(guan)聯的(de)主(zhu)題將在(zai)后面討論。


A系列海藻(zao)糖雙子的(de)水不溶(rong)性(xing)使我(wo)們能夠(gou)將其(qi)(qi)作為單(dan)分子膜放置在(zai)(zai)(zai)水亞相上。31-33借助(zhu)表面天平,雙子座單(dan)分子膜被可移動屏障緩(huan)慢壓(ya)(ya)縮(suo),在(zai)(zai)(zai)此過程中(zhong),監測膜壓(ya)(ya)力和面積(ji)。由此產生的(de)“壓(ya)(ya)力-面積(ji)等溫線(xian)”揭示了雙子分子在(zai)(zai)(zai)單(dan)分子膜中(zhong)的(de)堆積(ji)行為。圖(tu)1中(zhong)給出了A-12和A-18的(de)代(dai)表性(xing)等溫線(xian),其(qi)(qi)中(zhong)縱坐標是施(shi)加在(zai)(zai)(zai)單(dan)層上的(de)表面壓(ya)(ya)力(單(dan)位為mN/m或達因/cm),而橫坐標是在(zai)(zai)(zai)給定壓(ya)(ya)力下膜中(zhong)每個分子的(de)面積(ji)(單(dan)位為?2)。


圖1顯示了約120?2的(de)A-12和(he)A-18(以及此(ci)處未(wei)繪制的(de)中(zhong)間鏈長度)的(de)剝離區域(即,由于分子(zi)間接觸,等溫(wen)線(xian)偏(pian)離基線(xian)的(de)區域)。通過校準,烷基糖苷的(de)平均面(mian)積為49?2,34,35,而(er)烴鏈的(de)分子(zi)橫(heng)截面(mian)約為20?2。33圖l由A-12和(he)A-18幾(ji)(ji)(ji)乎相同的(de)等溫(wen)線(xian)支持的(de)最(zui)簡單(dan)解釋是(shi),海藻(zao)糖間隔基而(er)不是(shi)其碳氫(qing)化合物取代基主導界(jie)面(mian)填(tian)充。顯然,烷基從空氣(qi)/水界(jie)面(mian)投射到空氣(qi)中(zhong),對薄(bo)膜(mo)的(de)幾(ji)(ji)(ji)何要求幾(ji)(ji)(ji)乎沒有貢獻(xian)。盡管如此(ci),鏈間的(de)分子(zi)間范德華力(li)穩定了薄(bo)膜(mo)。這是(shi)因為崩塌壓(ya)力(li)(即單(dan)分子(zi)膜(mo)最(zui)終“斷裂(lie)”時的(de)高壓(ya))與鏈長正相關,例如A-18(53 mN/m)和(he)A-12(47 mN/m)。

當固體(ti)A-10和(he)(he)A-12雙子座浸(jin)入水中(zhong)(zhong),并在3-4小(xiao)時后通(tong)過光(guang)學顯微鏡進行檢查時,囊(nang)泡(pao)結構從表(biao)面(mian)發(fa)出(chu)(圖2)。當1毫米(mi)(mi)A-10的(de)(de)乳白色懸浮液(ye)通(tong)過100納(na)米(mi)(mi)聚碳酸酯過濾器多次(ci)擠出(chu)時,根據動態(tai)光(guang)散(san)(san)射,形成了直徑為100納(na)米(mi)(mi)的(de)(de)囊(nang)泡(pao)群。另(ling)一方面(mian),固體(ti)A-l4、A-16和(he)(he)A-18的(de)(de)水合(he)作用在室溫(wen)下(xia)不(bu)會產生(sheng)(sheng)囊(nang)泡(pao)。A-10/A-12與其更高同系物(wu)之間的(de)(de)行為差(cha)異表(biao)明,相態(tai)在其中(zhong)(zhong)發(fa)揮(hui)了作用。因此(ci),已(yi)知(zhi)短鏈(lian)脂質在室溫(wen)下(xia)存在于流體(ti)“液(ye)晶(jing)(jing)”相中(zhong)(zhong),而長鏈(lian)脂質更喜歡高度(du)有序的(de)(de)“凝(ning)膠(jiao)”相。36凝(ning)膠(jiao)到液(ye)晶(jing)(jing)的(de)(de)轉化發(fa)生(sheng)(sheng)在轉變溫(wen)度(du)Tm。在Tm上方,水可以進入“松散(san)(san)”液(ye)晶(jing)(jing)晶(jing)(jing)格的(de)(de)雙層中(zhong)(zhong),從而在水合(he)時形成囊(nang)泡(pao)和(he)(he)小(xiao)管。即使在室溫(wen)下(xia),A-18U(即9位順式不(bu)飽和(he)(he)的(de)(de)A-18)的(de)(de)水合(he)作用也會產生(sheng)(sheng)大量的(de)(de)囊(nang)泡(pao)和(he)(he)小(xiao)管(圖2),正如人們(men)從一種Tm無疑(yi)遠低于室溫(wen)的(de)(de)化合(he)物(wu)中(zhong)(zhong)所預期的(de)(de)那樣。

差示掃描量熱(re)法掃描是通過在(zai)量熱(re)計中(zhong)以10°C/h加(jia)熱(re)2 mg/mL gemini懸浮液獲得的(de)(de),其吸(xi)熱(re)峰(feng)分(fen)別對應于(yu)a-14、a-16和(he)a-18的(de)(de)Tm值33、48和(he)61°C(圖3)。只有A-l8有一個尖(jian)峰(feng),表明在(zai)緊密(mi)組織的(de)(de)分(fen)子陣(zhen)列中(zhong)協同熔(rong)化。當固體A-14和(he)A-16在(zai)45°C下水(shui)合(he)時,觀察到與(yu)圖2中(zhong)類似的(de)(de)光學顯微鏡圖像,而(er)不是在(zai)室溫下缺乏雙層形成。

迄今(jin)為(wei)止的總結是:A系列海(hai)藻(zao)糖雙子(zi)(zi),對(dui)于10或更長的鏈不(bu)溶于水,形成單分子(zi)(zi)膜,其(qi)界面區域主要(yao)由糖間(jian)隔基控制(zhi)。分子(zi)(zi)間(jian)的鏈/鏈相互作用穩(wen)定(ding)了薄膜。A-8在水中形成小(xiao)膠(jiao)束(shu),而(er)水合(he)(he)后,更高的同系物聚集(ji)成泡狀(zhuang)和管狀(zhuang)結構,在光學顯微鏡(jing)下可見。水化性質與凝膠(jiao)到液晶的轉變相一致,事實上,轉變溫(wen)度(du)可以通過量熱法確定(ding)。因此,海(hai)藻(zao)糖雙子(zi)(zi)的行為(wei)與磷脂(zhi)和合(he)(he)成雙鏈表面活性劑的行為(wei)相似。17,37就好(hao)像海(hai)藻(zao)糖不(bu)是雙子(zi)(zi)座的間(jian)隔物,而(er)是一個特(te)別大但傳統的頭(tou)群。


值得注意的(de)是,我(wo)(wo)們的(de)雙(shuang)子(zi)座研(yan)究利用了許(xu)多技術(shu),包括合成、薄膜研(yan)究、光散射、顯(xian)微鏡、量熱法(fa)(fa),以(yi)及(我(wo)(wo)們很快就會(hui)看到)張量法(fa)(fa)、電導法(fa)(fa)和(he)分子(zi)力學。在膠體化學中,一種平衡的(de)觀(guan)點不能僅僅通過(guo)一種方法(fa)(fa)來實(shi)現(xian),而只能通過(guo)多種方法(fa)(fa)之間(jian)的(de)一致性來實(shi)現(xian)。


B系列海藻糖雙子座


接下來,我(wo)(wo)們(men)提(ti)出了(le)(le)在海藻糖衍生物(wu)上(shang)放置陽離子基團以改善其水(shui)溶性的(de)想法。必須承認,我(wo)(wo)們(men)對更易管理的(de)水(shui)溶性化(hua)合(he)物(wu)的(de)前景感到一定程度的(de)高興。B系列就這(zhe)(zhe)樣出現了(le)(le)。然而,我(wo)(wo)們(men)立(li)即(ji)面臨新的(de)問題。雖然B-12和B-14是作為純(chun)化(hua)合(he)物(wu)獲得(de)的(de),但B-16和B-18并非如此。后兩種(zhong)Gemini的(de)產率較低,提(ti)供了(le)(le)令人(ren)滿意(yi)的(de)光譜(pu)數(shu)據,但它們(men)的(de)元素分(fen)析偏(pian)(pian)離了(le)(le)理論1-2%(這(zhe)(zhe)使我(wo)(wo)們(men)懷(huai)疑(yi)存在無機(ji)雜質(zhi))。無論如何,由于B-16和B-18不符合(he)我(wo)(wo)們(men)的(de)高純(chun)度標準(zhun),我(wo)(wo)們(men)在注(zhu)意(yi)力上(shang)輕(qing)視了(le)(le)它們(men),而偏(pian)(pian)愛(ai)它們(men)的(de)矮表親。

實驗段
